Limitations of restriction fragment length polymorphism (RFLP):
- Requires large amounts of sample DNA
- Can be slow and cumbersome
- No potential for automation
- Few loci detected per assay
- Low levels of polymorphism in some species requiring detection by radioisotope
- Single-copy probes can be time-consuming
- Requires a suitable probe library
- Requires the combined process of DNA fragmentation, probe labeling, electrophoresis, hybridization, washing, blotting and autoradiography
