Both techniques involve the separation of nucleic acid molecules based on their size using gel electrophoresis. After gel electrophoresis, the separated nucleic acid molecules for both types are blotted onto a solid support membrane using capillary transfer. They also involve the use of a radiolabel enzyme as their probe labels. Additionally, the labeled nucleic acid probes hybridized to the target sequences are detected using the same methods, including autoradiography (for radioactive probes), chemiluminescence, or colorimetry.
