High Throughput Screening (HTS)
AAT Bioquest's Screen Quest™ high throughput screening (HTS) platform offers optimized assay kits compatible with 96-well and 384-well plate formats, enabling efficient compound screening with minimal hands-on time. Targets of interest include calcium and cAMP, as well as membrane transporters and ion channels, to accelerate drug discovery and GPCR research.
Our HTS Technologies

Screen Quest™ — Ready-to-use assay kits optimized for 96/384-well HTS formats
Fluo-8® — 4x brighter than Fluo-4 for improved calcium detection sensitivity
Calbryte™ — Next-generation calcium indicators with enhanced cell loading and retention
cAMP Biosensor Cell Lines — Stable lines with FRET-based cAMP sensors for real-time GPCR monitoring
For individual calcium indicators and ion probes, explore our Physiological Indicators product lines.
cAMP Assays

cAMP Assays provide sensitive detection of intracellular cAMP levels for studying Gs and Gi-coupled GPCR signaling.
  • Homogeneous TR-FRET and fluorescence polarization formats
  • No-wash protocols minimize well-to-well variability
  • Compatible with adherent and suspension cell types
  • Suitable for agonist, antagonist, and inverse agonist screening
Calcium Assays

Calcium Assays detect intracellular calcium mobilization, the primary readout for Gq-coupled GPCR activation.
  • Screen Quest™ Fluo-8® delivers 4x brighter signal than Fluo-4
  • Calbryte™ indicators with superior cell retention and no dye leakage
  • Probenecid-free formulations eliminate transporter interference
  • No-wash protocols for simplified HTS workflows
  • Orange/red indicators (Calbryte-590, Rhod-4) for multiplexing
Transporters

Transporter Assays measure activity of glucose transporters, fatty acid uptake proteins, and ABC drug efflux pumps for ADME and drug resistance studies.
  • Non-radioactive glucose and fatty acid uptake detection
  • MDR/P-glycoprotein efflux pump activity assays
  • Chloride channel and ion transporter kits
  • HTS-compatible 96-well and 384-well formats
Other HTS Assays

Other HTS Assays address diverse screening needs including GPCR desensitization, electrophysiology, and antimicrobial resistance.
  • Beta-arrestin translocation for GPCR activation monitoring
  • Membrane potential assays with voltage-sensitive dyes
  • Beta-lactamase inhibitor screening for antibiotic resistance research
  • Real-time kinetic and endpoint measurement options
