What are the advantages of using synthetic oligonucleotide probes for performing in situ hybridization?

Posted March 27, 2024

Answer

One advantage of synthetic nucleotides is that they are cost-effective compared to other probe types. Synthetic oligonucleotides are also readily available and relatively easy to work with. Synthetic oligonucleotides (particularly shorter probes) can penetrate tissue samples more efficiently than larger DNA fragments or RNA probes. Additionally, they offer excellent reproducibility in FISH experiments and can be designed with high sequence specificity to target complementary DNA or RNA sequences. They are also resistant to degradation by RNases.
