One of the best hybridization buffers to use for DNA hybridization is SSC (saline-sodium citrate) buffer. Addition of SDS (sodium dodecyl sulfate) to the buffer is also highly beneficial as SDS prevents non-specific binding. Alternatively, Church & Gilbert’s hybridization buffer, which is a sodium phosphate buffer that also includes SDS, is also an excellent buffer to use.
