How do I reduce sample exposure in order to minimize photobleaching?

Answer

In order to minimize photobleaching, one should reduce the amount of sample exposure to light. This can be done by focusing the image using transmitted light, focusing on the region next to the area of interest, or through using substandard exposure time. One should also store fluorophores in a dark space, incubate samples in the dark, and turn off the microscope’s light between images. If a specimen requires mounting, antifade mounting media may be used to reduce fading.
