BES-Buffered Saline (2X) (0.05 M, pH 6.95) Preparation and Recipe
BES-buffered saline is often used in the calcium phosphate-mediated transformation of mammalian cells.
To prepare L of BES-Buffered Saline (2X) (0.05 M, pH 6.95):

Sodium Phosphate Dibasic (mw: 141.96 g/mol) | 0.21 g | 0.0015 M |
- Prepare 800 mL of dH2O in a suitable container.
- Add 10.66 g of BES to the solution.
- Add 16.36 g of Sodium chloride to the solution.
- Add 0.21 g of Sodium Phosphate Dibasic to the solution.
- Adjust solution to desired pH using 1M NaOH (typical pH = 6.95)
- Add dH2O until the volume is 1 L.
