Glycine-HCl Buffer (0.1 M, pH 3.0) Preparation and Recipe
This low-pH buffer is used extensively with affinity chromatography, particularly with proteins and antibodies. It is also used for cell culture and other biological applications.
To prepare L of Glycine-HCl Buffer (0.1 M, pH 3.0):

Hydrochloric acid (mw: 36.46 g/mol) | 0.832 g | 0.02 M |
- Prepare 800 mL of distilled water in a suitable container.
- Add 7.5 g of Glycine to the solution.
- Add 0.832 g of Hydrochloric acid to the solution.
- Adjust solution to final desired pH using HCl or NaOH
- Add distilled water until the volume is 1 L.
