SSC (Northern Blot, 20x) Preparation and Recipe
SSC (saline sodium citrate) buffer is commonly used in Northern blotting as a hybridization or wash buffer.
To prepare

Sodium chloride (mw: 58.44 g / mol)##
Trisodium citrate (mw: 258.07 g / mol)##
  • Prepare 800 mL of distilled water in a suitable container.
  • Add # of Sodium chloride to the solution
  • Add # of Trisodium citrate to the solution
  • Add distilled water until the volume is 1 L.
  • Adjust the pH to 7.0 with NaOH or HCl. Autoclave the solution and store it at room temperature. It will last for 1 yr or longer.
