SSC (Northern Blot, 20x) Preparation and Recipe

SSC (saline sodium citrate) buffer is commonly used in Northern blotting as a hybridization or wash buffer.

To prepare

L of SSC (Northern Blot, 20x):

Table 1. Required components

Component	Amount	Concentration
Sodium chloride (mw: 58.44 g / mol)	#	#
Trisodium citrate (mw: 258.07 g / mol)	#	#

Prepare 800 mL of distilled water in a suitable container.
Add # of Sodium chloride to the solution
Add # of Trisodium citrate to the solution
Add distilled water until the volume is 1 L.
Adjust the pH to 7.0 with NaOH or HCl. Autoclave the solution and store it at room temperature. It will last for 1 yr or longer.
